We also showed that only definitive myeloid cells, but not primitive, rely in this two-hit system for their differentiation. That helped dissect how 2 embryonic Mac populations respond differently to tissue injury and which one promotes regeneration.

But when we added both hits, we successfully initiated myeloid differentiation in a human erythroleukemia line. Jak2/Stat3 are hyperactive and targets in many hematopoietic malignancies. GRN supplementation could enforce their differentiation and help eradication.

Until the #zebrafish helped us identify the mechanism! 🐟 Grn synergizes with Jak2/Stat3 to drive myeloid determination. It is a two-hit system. If GRN is not present, or Jak2/Stat3 is inactive, myeloid precursors fail to differentiate.

The role of progranulin (GRN) in human #hematopoiesis was a mystery. However, it is the most abundant transcript in human Macs and granulocytes!! We tried for years to discover its hematopoietic function, but failed.

Congratulations!! 🎉🎊 Very interesting work! Do you know if EMPs (pubmed.ncbi.nlm.nih.gov/17959717/) or EMP-derived Mac’s accumulate at the injury site? They both would be Gata1+. It would be interesting if EMPs migrate to provide a localized source of myeloid cells at the injury site.
