Did you know that just 100-200 gold particles in your #immuno-EM data can give you an incredibly good idea of the #labelling distribution and reveal positive signals that you may not immediately recognise? It's amazing how reproducible this " #100-gold-method" is. #stereology doi.org/10.1369/jhc....

#Correlative Light and Electron Microscopy, #CLEM, is a modern #method combining light and #electronmicroscopy data. Although it is considered “new”, it was already used in the early 1960s, for instance to study the #ultrastructure of mitotic cells in this beautiful example: doi.org/10.1083/jcb....

The toxic, #arsenic -containing #Cacodylate buffer is still used in many #electronmicroscopy labs today. Although it is a good buffer, it is no longer needed today as there are better and safer alternatives that perform just as well - #PHEM buffer (Schliwa, 1981) for example. doi.org/10.1111/j.13...

I'm gonna piss a few nerds off but this is an example of why a lightsheet microscope should be used to study organoids. The detection optics are great for imaging in XY but they suck axially. Sure, yeah, hocus pocus, RI matching, correction collar, blah blah, but it's physics, baby.

We are rather skeptical about conclusions based on #AI-driven / #deeplearning #image #analysis (not towards an application as a tool). This viewpoint is supported by a recent study on medical imaging, which we believe can be applied to #bioimage data in general. #microscopy doi.org/10.1038/s415...

It's truly surprising that the #tokuyasu method is so underutilized in #fluorescence #microscopy! It eliminates the need for permeabilisation and increases Z resolution to 50nm on any microscope. It is also the method used in the Nobel Prize-winning work on #super-resolution. doi.org/10.1177/0022...

It is truly remarkable that Fritiof Sjöstrand was able to draw such precise conclusions about the cell in the era of primary osmium tetroxide #fixation, preceding the introduction of glutaraldehyde by Sabbatini in 1963. As we recently posted, our preferred #fixation #buffer for mammalian cells and most tissues is PHEM. Marine invertebrates, especially #plankton, seem to fix better with a variation, as shown in this publication by Montanaro. Of course, osmolarity has to be considered as well. doi.org/10.7717/peer...
