Sad news. I saw the Frames a few times, although not enough. The first at a small gig in around 1999. Hadn’t heard a single song before the show, bought their two (at the time) albums immediately after.

Many scientists I've been talking to are freaking out about NIH "immediate deposit" mandate and the publishers, like journals in the Nature family, telling them they have to pay ~13k to satisfy the mandate. People have been paying it - but they don't need to.

Yeah this. Understand the drive to “protect children” but there are valuable uses for these platforms - organising community action and events, music and sport - which also get lost. and if you ban teenagers from Snap etc - they go to more unregulated sites, chat rooms etc… I don’t have an answer

It is the anniversary of the My Lai massacre today. I'd say that even more than most years, it's important to remember what happened. And the role Hugh Thompson, an active serviceman, played in both stopping and publicising it. Despite enormous military, political and public pressure to stay quiet.

Using copilot to reformat NMRs and check count the number of hydrogens. It’s like dealing with a lazy teenager. After the first few it starts cutting corners and not counting properly and I have to give it a telling off. Why has copilot been programmed to be like a shoddy coworker? #chemsky

#ChemSky - any experience with different models / brands of centrifugal evaporator (Labconco, Genevac, Speedvac, others…) ? We’re doing more plate and vial based parallel chem and evaporation is often the bottleneck. I last used a Genevac in about 2013 so I am out of date…other suggestions welcome!

So us medicinal chemists do occasionally pay attention to our process chem buddies and we know NaH in DMF is a bit explody on occasion. But we also make a lot of insoluble things. Are THF / DMF mixtures OK…? how much DMF makes it a problem? #chemsky

Target Engagement Assays in Early Drug Discovery
In target-based drug discovery, quantification of target engagement is required to build structure–activity relationships and develop a potent clinical candidate. Target engagement data also provides evidence of a drug’s mechanism of action (MoA) which although is not required for approval, can increase the chance of a successful clinical outcome. Consequently, a plethora of assays has been developed to provide information about target engagement on isolated proteins and in cells. These techniques monitor changes in stability, structure, optical properties or mass difference between proteins and their complexes with ligands. They also provide characterization of the compound with thermodynamic, kinetic and structural binding parameters. The diversity of approaches reflects the challenges faced when drugging different protein classes, with each method having advantages, trade-offs and target specificity.
