Doing proteomics on different yeast biofactories showed us that strains from different origins respond differently to the stress caused by recombinant protein expression—something I had not expected. link.springer.com/article/10.1...

Interestingly, we also see that additional pathways remain to be uncovered, as BAG6 knock down only partially stabilize misfolded proteins and degrons, highlighting to potential high redundancy in cytotolic protein quality control

Using BioID, we surveyed multiple misfolded cytosolic proteins and observed broad chaperone recruitment, including BAG6 in some cases. RNAi confirmed BAG6 and RNF126 are required for efficient turnover of several mutants.

BAG6 mediates the localization of the Tail anchored membrane proteins via the GET pathway and target for degradation mislocalized transmembrane proteins as well as aberrantly translated proteins– summarizing 20 years of work from the Kawahara, Hegde, Ye, Hartl, Wu and Elledge labs to name a few.

I survived the 2025 EMBO protein quality control meeting - it was amazing despite a defective projector on day 1, and epileptic crisis on day 2, a black out in day 3, and we all got a shaky wake up call ~6am on day 5 by an earthquake… never a dull moment but most importantly great science

One limitation of our study is the use of rapidly dividing cells, where the protein homeostasis network likely differs from that in differentiated cells with lower protein production. Despite our efforts, we have yet to identify the E3 ligase involved—but we’re not giving up, so stay tuned!

In this case, the disease-associated mutation increases interaction with the DNAJA2 chaperone, which appear to be dispensable for the folding of the wild-type protein. This interaction delays the proteolysis of the mutant protein, potentially buffering its deleterious effects.

Using BioID, we identified chaperones, including the co-chaperone DNAJA2, that interact more with the unstable thiopurine S-methyltransferase (TPMT) variant than the wild type. DNAJA2 deletion accelerates degradation of the mutated TPMT but does not affect the wild-type protein

Many disease-associated mutations are missense mutations causing single amino acid changes that signal proteins for degradation. We surveyed 18 unstable variants from 13 cytosolic proteins and found that all but one are at least partially degraded via proteasome- and ubiquitin-dependent pathways.

When I was on the bench It wasn't uncommon for experiments to fail, then fail again, and again (you probably know the feeling). A rejected grant is not that different In most cases, it’s not the fall that matters but how we bounce back. Each trial makes us more experienced; well that’s the theory 😉
