The switch runs through a protein called ERRγ. Blocking it in cultured muscle cells shut the switch down completely. Mice lacking cardiolipin in muscle also took up much more glucose but did not simply burn it for energy. They shunted much of it into pathways that support antioxidant defenses.

So why does muscle respond this way? It is a defense mechanism. Strained mitochondria produce more reactive oxygen species (ROS), which can damage cells but also act as a signal. An antioxidant blunted the fiber switch in cardiolipin-depleted muscle cells.

When the scientists lowered cardiolipin in young mice, mitochondrial structure and function broke down and the muscle shifted toward slow-twitch fibers, as it does in aging humans. Restoring cardiolipin to roughly 2/3 of normal began to reverse muscle wasting and prevented the early deaths entirely.

The answer might lie in cardiolipin, a fat molecule found almost exclusively in the inner membrane of mitochondria. It falls with age in mouse muscle and tends to fall in human muscle.

As we age, muscles weaken and their mitochondria work less well. Yet muscles also shift from fast-twitch fibers, built for power, toward slow-twitch fibers, built for endurance and richer in mitochondria. Why would aging muscle lean harder on the very machinery that is failing?

It's out now in Nature Metabolism 💉 Most obesity meds hit 1-2 hormone receptors (GLP-1R ± GIPR), and they're quite effective, but doses a being pushed to achieve greater weight loss = more side effects. We combined retatrutide (GLP-1R/GIPR/GCGR) with cagrilintide (AMLNR/CALCR) in obese rats.

A fuel partitioning perspective on appetite suppression by GLP-1 receptor agonists
Appetite suppression by glucagon-like peptide-1 receptor agonists (GLP-1RAs) is thought to result from direct actions on neural circuits controlling food intake. In this article, we (1) propose that a shift in fuel partitioning toward fat oxidation caused by GLP-1RAs contributes to appetite suppression and (2) explore potential lines of research to test this hypothesis.
