3/4 We further uncovered luminal γ-tubulin ring complexes (γ-TuRCs) anchored to the MT wall via the augmin complex — candidate nucleation sites for central pair (CP) biogenesis! we also surprisingly found tube-like densities underneath the luminal ring which may be the selective gate?

2/4 Resolving native BBs at subnanometer resolution in situ, we identified 11 ciliary-base microtubule-associated proteins (CMAPs, including 5 novel core components) specifically orchestrating the triplet-to-doublet transition along the proximal-distal axis. 🔬

In situ architecture of the basal body reveals regionalized assembly and central pair microtubules nucleation
The basal body (BB) is essential for axoneme assembly and ciliary function, and its defects are associated with ciliogenesis disorders and ciliopathies (1,2). However, the molecular basis of the BB's ...

First day here and I brought a paper. Three years of cryo-FIB-ET on human airway cells, nine proteins identified and two new PCD genes at the end of it. Grateful to the teams in Munster (Omran Lab), Boston (@alanbrownhms.bsky.social, Hawkins Lab) and Geneva (@centriolelab.bsky.social).

The results here re 3D classification are quite impressive. Would be interesting to see how much the LPP improves classification of near-native samples with extreme compositional heterogeneity (e.g. derived from cellular fractions)

Laser phase plate improves structure determination of small proteins by cryo-EM
Phase plates can in principle overcome the poor image contrast in electron cryo–microscopy (cryo-EM) and the resulting limits on the structural reconstruction of small proteins. However, previous desi...

Always looking for ways to improve cryo-ET lamella quality. In last week’s #journalclub, we discussed what seems to be a promising and practical in-chamber sublimation approach for mitigating ice contamination and curtaining during lamella prep. Preprint-- www.biorxiv.org/content/10.6...
