We developed Adaptive Polishing for the consistent preparation of ~150 nm thin FIB-lamellae. This was done by using imaging feedback to adjust parameters for each lamella, or Adaptive Milling, rather than applying a fixed recipe. Preprint: doi.org/10.64898/202...

Single-cell visual proteomics of a minimal bacterium reveals structural coordination of gene expression machineries
Cryo-electron tomography (cryo-ET) resolves and quantifies an almost complete view of bacterial translation, revealing how gene expression machineries coordinate across the processes of transcription,...

🎉 Out today in Cell: the paper I've been looking forward to sharing for a long time 🎉 We used cryo-ET to find a molecular machine nobody knew existed on the surface of a minimal bacterium, and worked out what it does. 🧵(1/7) #TeamTomo #cryoET

Releasing TOMATO, the TOMogram Annotation TOol! A lightweight Python GUI for going through hundreds of cryo-ET tomograms. Flag what's in each one, grade its quality, and end up with a table you can filter and act on. github.com/alkemasimon/...

With Cryo-ET/EM, the macromolecules we study (and biological questions we ask) are SNR-limited. Here, we share Nilas, a low-voltage milling method named after thin sea ice. Nilas produces thin (sub 50 nm in places!), minimally damaged lamellae. biorxiv.org/content/10.64898/2026.07.21.739890v1

Optimized cryo-FIB milling strategy to generate thin, minimally damaged biological lamellae
Focused ion beam (FIB)-milling has been adapted to thin frozen cells for visualization of macromolecular structures in situ with cryogenic electron microscopy. However, only a few large and abundant c...

In a series of 3 papers and preprints, we’re thrilled to share with you the working laser phase plate. In collaboration with research led by Holger Müller at UC Berkeley, this is a huge innovation in imaging to make small and faint objects inside cells visible. bit.ly/4vK9LVn

New preprint🧊🔬Introducing LowDoseWizard—a guided SerialEM workflow for rapid, standardised cryo-TEM setup! Setting up low-dose can be slow & daunting, especially for new users. LowDoseWizard automates optics, camera, and alignment steps from minimal input — full setup in <15 min. #cryoEM #SerialEM
