(1/4) We are very excited to share our latest preprint from @sauer-lab.bsky.social: Mega Expansion Microscopy (Mega-ExM)! We combine iterative hydrogel expansion with whole-proteome NHS labeling and post-expansion immunostaining, reaching tunable expansion factors up to ~1500× in whole cells 🤯

Exciting new preprint from Ignacio Vega Vásquez, Sauer lab, my team and collaborators: #Mega-ExM. From just ~20 NPCs imaged with a 10× objective on a confocal, we reconstructed the 3D structure at ~33 Å. A step toward optical structural biology in intact cells. www.biorxiv.org/content/10.6...

🚨New paper out today in @acs.org ACS Nano, showing how expansion microscopy can visually quantify the cell surface receptor turnover mediated by signalling & receptor organising machineries on the surfaces of endosomes, some of the smallest organelles in cells. 🧪 🔬 1/ pubs.acs.org/doi/10.1021/...

But none of our existing imaging methods (SoRa, STORM…) could really visualize integration within the bundled microtubules in neurites. So Ciarán set up U-ExM expansion microscopy, then performed it on microinjected neurons - now we could see plus-end growth and in-lattice renewal everywhere! 🧵 7/9

The images shows an array of 6 Expansion microscopy images of cell nuclei (three columns and 2 rows). each image was taken with a different SPY-DNA probe to stain the nuclear DNA and is pseudocolored to the  fluorescence wavelength of the SPY-DNA probe. Trom top left to bottom right: SPY505-DNA (green); SPY555-DNA (magenta); SPY595-DNA (yellow); SPY620-DNA (orange); SPY650-DNA (red); SPY700-DNA (darker red). a white scale bar of 50 micrometers is given for each image to get an impression of the scale of each nuclei.
