Dendritic cells (DCs) can acquire distinct functional states. To examine how different DC states orchestrate anti-tumor immunity, Koufaki et al. develop mouse models to label and ablate CCR7+ DCs and find that this activation state is essential for tumor control by uniquely priming tumor-specific cytotoxic T lymphocytes (CTLs), sustaining their function and enabling responses to immunotherapy.

Respiratory infection and cardiovascular disease risk are tightly intertwined clinically, but the mechanisms of this relationship are largely unknown. Here, Downey et al. reveal how influenza traffics to the heart from the lung largely within pro-DC3s, which unleashes a damaging type I interferon program that compromises cardiac function.

Eomesodermin+ CD4+ T cells are critical for curative immunotherapy outcomes
Type-1 regulatory T cells (Tr1) promote immune tolerance during chronic infection, autoimmunity, and transplantation, but the role of Eomes in Tr1 differentiation and function after cell therapy is unclear. Zhang et al. find that Eomes controls the acquisition of regulatory and cytotoxic function in CD4+ T cells that are critical for limiting immune toxicity, while maintaining persistence and tumor control after immunotherapy.
