#FluorescenceFriday - experiments take so long (especially for PIs that moonlight as postdocs), between breeding, tamoxifen, waiting for the 🐭 to grow up, slicing, clearing, staining for CD206 then finding a chunk of time on the 🔬...but then I get to image all my fav cells & it's SO worth it. ❤️

I did well in my Men's NCAA basketball bracket this year, picked the Final 4 & winner Michigan. Turns out picking teams because 'there's good science being done there' or 'I like their mascot' or 'they invited me for a talk' or 'my good friend went to school there' is not a bad way to do it. 🏀🔬🧪🧫🧠

Fibroblasts underpin immune-competent niches in the healthy and diseased CNS. Schematic representation of the topological organization of fibroblasts along the vascular tree and across meningeal and brain parenchymal compartments in the context of (A, B) homeostasis, (C, D) infection, (E) autoimmunity, and (F) sterile tissue injury. The provision of cytokines, chemokines, survival factors, and ECM components by local fibroblasts are indicated in each condition. The legend of depicted cell types and vasculature is provided. (A) In the homeostatic CNS, fibroblastic cells include mural cells, perivascular fibroblasts situated abluminal to mural cells, and meningeal fibroblasts. (B) In the steady-state condition, PDPN+ FRC-like cells are limited to DALTs positioned at the intersect of rostral-rhinal, olfactory, and superior sagittal sinuses. In (C) chronic dural infection and (E) autoimmunity, persistent inflammation is associated with TLS formation. TLS are underpinned by FRC-like cells and FDCs providing T and B cell chemoattractants, survival factors, and canonical markers of FRCs. (D) In infections of the brain parenchyma, similar chemokines and FRC markers are upregulated in the absence of TLS formation. (E) In the context of deep gray matter injury, perivascular fibroblasts of the Virchow-Robin space constitute a major source of scar-forming cells. In addition to profuse ECM deposition, scar-forming fibroblasts secrete myeloid cell survival factors and chemokines to drive early and late phase scarring, respectively.

#FreehandFigureFriday continues...this from a figure I mocked up for a review but ended up on the 'digital' cutting room floor - scanning EM of the meninges of large mammals like dogs (this from PMID: 1094039) is really great, gives a great appreciate for 'smoothness' of SAS surfaces.

Sophia did an AMAZING job with this project - big props for innovating on 🔬 & image analysis - the first figure of this paper took a bit because she had to develop tools to study arachnoid barrier functional integrity in pups & quantify Cldn11 junctions [we couldn't just say 'they look weird' 😂] 3/5

"Biomedical and life science articles by female researchers spend longer under review" from @plosbiology.org. After I read this sentence, I counted & I have 43 papers w/ a female first and/or last author. Lots of time wasted in longer reviews/revisions. 😕 journals.plos.org/plosbiology/...
Even though, for each manuscript, time under review is only 7–15 days longer for female- than for male-authored articles (Fig 2), accumulated over a woman’s career, these delays can be very substantial. For instance, for every 50 papers published by a female author, she will have spent on average 350–750 days longer than her male counterparts waiting for reviews and editorial decisions and/or revising manuscripts.

Clark has a had a rough couple weeks...'mom' has been too busy writing grants to hike ⛰️...he was bit by another dog on a walk [required a vet visit & staples 🥴]...BUT 'mom's' grant is submitted, wound is healed & it's a GREAT day for a long hike....made perfect with a dip in the creek at the end. ❤️

In the normal leptomeninges, meningeal fibroblasts are present and associate with meningeal blood vessels. In the presence of medulloblastoma (MB) metastatic tumour cells, secreted platelet-derived growth factor (PDGFA) ligands in the local microenvironment chemotactically recruit meningeal fibroblasts. Meningeal fibroblasts are reprogrammed to become MB-MAFs, expressing distinct transcriptomes and secretomes, including bone morphogenetic proteins 4 and 7 (BMP4/7). Active BMP and PDGF signalling further enhance both the colonization of the leptomeninges by MB cells as well as promoting the growth of established metastases.

Great new work led by @biomarina-vg.bsky.social on zebrafish meninges! Particularly 🤩 is the descriptions of ependymin-expressing meningeal cells (not a fibroblast!) & ablation studies showing their essential role in viability. So much more to learn! Congrats Marina & team on this great work! 🦓🐟🧠

I was imaging this weekend, took this image because it was beautiful to me, the blood vessels, fibroblasts, astrocytes, & macrophages as steadfast guardians at the brain's edges. Cheers to the 'guardians' of all that is important. ❤️
