Thanks a lot, Mike! Definitely DyeCycling can be realised with other reversible chemistries and we work on that. But for FRET resolution at the nanometer level, the linkers should be short. Hence, less sure about HaloTag & Co... May work in some cases. Curious to hear your ideas.

Right in time for #smFRET’s 30th anniversary, DyeCycling enables true single-molecule kinetic analysis, revealing hidden heterogeneities and even time-variant kinetic changes of individual molecules.

The trick is to harness reversibly binding fluorescent probes – and their cumulative photon budgets – rather than single dyes whose bleaching terminates the observation immediately. High SNR is retained through efficient nanophotonic background suppression, demonstrated using DNA & protein systems.
