aCOS is a spectroscopy-based technique to determine affinities between molecules, e.g. small molecules and targets, or between two or more interaction partners leveraging the change of complementary fluorescence spectroscopy parameters upon binding or dissociation.

This was a new adventure for us to develop an assay to stress bacteria 🧫 and test how good they adapt to antibiotic stress. We stress bacteria 🦠 with e.g. Ciprofloxacin and see if they adapt via a provided mobile integron system ± peptide. We determine many factors using also microscopy… 5/

DNA elements were important for mechanics, but also protein mutations decreased recombination significantly. In particular the C-terminal extensions (the little pig-like ;) tails linking IntI1) stabilised the complex significantly and truncation reduced recombination. Hm💭 - could we maybe … 2/

At least 10% of the V. cholerae integron cassettes have novel antiphage activities (see below). Considering that there are 25,000 distinct cassettes of unknown function, integrons constitute an extraordinary and almost inexhaustible reservoir of antiphage functions!
