H1 blockers may also affect inflammation through NF-κB "Desloratadine inhibits constitutive and histamine-stimulated nuclear factor-kappaB activity consistent with inverse agonism at the histamine H1 Receptor " pubmed.ncbi.nlm.nih.gov/38953634/
@manruipa
24/35

And here is another interesting feature of H1 antihistamines that goes beyond simply preventing histamine from binding to the receptor.

The H1 receptor has some constitutive activity: it can maintain intracellular signalling even in the absence of histamine.

One pathway it can activate is NF-κB, a major regulator of inflammatory gene transcription.

Many H1 antihistamines actually behave as inverse agonists. That means they do not merely prevent histamine from activating H1; they also stabilise the receptor in a less active state and can reduce some of its basal signalling.

H2 blockers like famotidine and loratadine used in Long Covid can improve some cognitive and neuropsychiatric measures and an additional reduction of approximately 1.48 points on the Fatigue Assessment Scale. But long term, H2 blockers affect stomach acid production, B12 absorption and gut biome.
@manruipa
14/35

Famotidine alone has also been studied.

In a randomised trial of 50 patients with post-COVID cognitive impairment, 12 weeks of treatment was associated with improvement in some cognitive and neuropsychiatric measures.

And in 2026 came STIMULATE-ICP, a large multisite, open-label randomised controlled trial.

The pharmacological trial included 778 people with Long COVID. Of these, 193 received famotidine 40 mg + loratadine 10 mg daily for 12 weeks.

The combination produced an additional reduction of approximately 1.48 points on the Fatigue Assessment Scale compared with receiving no study drug.

The difference was statistically significant but small, and 12 weeks after treatment ended, the difference between groups was no longer significant.

"A tissue focus capable of maintaining or replenishing viral RNA and proteins could provide antigenic stimulation for months or years without causing major viraemia." One infection can burden you for years. Multiple, repeated infections? For the rest of your(shorter than expected) life.
@manruipa
10/35

So I do not think the most useful distinction is simply:

“reservoir” vs “debris.”

There is growing evidence that, at least in some patients, viral material is maintained inside tissues for extraordinarily long periods.

The more interesting question becomes what is happening inside that reservoir: whether there is continuous low-level replication, intermittent activity, transcription and antigen production without completion of every viral replication cycle, or specific cell types that allow persistence.

Demonstrating any of these is not the same as demonstrating abundant continuous production of fully infectious virions.

But immunologically, that may not even be necessary.

A tissue focus capable of maintaining or replenishing viral RNA and proteins could provide antigenic stimulation for months or years without causing major viraemia.

@envidreams.bsky.social I also have had heart palpitations, now diagnosed w/DAO deficiency and dust mite allergy(yay, I can justify my respirator w/something other than preventing infections!).
@envidreamz
The most honest Long Covid thread on the internet is currently happening inside a perimenopause Facebook group.

Woman posts she’s having palpitations and feels like she’s having a heart attack.

Then the group starts confessing:
“We are walking dead”

“just diagnosed with atrial fibrillation, I need a heart ablation”

“had to wear a heart monitor for 3 days”

“POTS driven by mast cells”

“constant flight or fight”

“autonomic testing at a POTS clinic”

“2 week holter found 2 arrhythmias”

“It’s worse when I drink alcohol”

“I lived it almost felt like dying..check your ferritin. It might also be because of inner ear crystals.”

Doctors stay clueless. They gaslight no matter what.

Covering Long Covid up with perimenopause should be illegal.
How do we report this and force it to stop?
HRT on a damaged heart is how people get a heart attack or stroke.

Senator calls for criminal investigation of RFK Jr after Guardian report Newly obtained records indicate health secretary lied about Samoa trip during Senate confirmation hearings last year
A senior Democratic senator is calling for a criminal investigation into the US health secretary, Robert F Kennedy Jr, following reporting by the Guardian that indicated he lied during Senate confirmation hearings last year.

Ron Wyden of Oregon said: “RFK’s platform is built on lies and grifts that leave a trail of dead children in their wake. There are consequences for lying to Congress.”

The Guardian’s reporting was based on letters that were obtained – by this news outlet and the Associated Press – following a Freedom of Information Act lawsuit.

Wyden said in a statement to the Guardian that he would be pressing Republican colleagues, who currently control the Senate, to issue a criminal referral of Kennedy to the Department of Justice for a criminal investigation. “And when Democrats retake the majority in the Senate, RFK will be at the top of the list of Trump officials the finance committee will hold accountable for endangering American families,” he said. Wyden currently serves as the top Democrat on the finance committee.
