Human megakaryocytes were found to have 2 SEL1L protein isoforms that increase in expression during megakaryopoiesis, although only 1 isoform was delivered to mature platelets. Studies using inducible mouse and constitutive zebrafish KOs demonstrated that SEL1L is necessary for efficient platelet or thrombocyte (fish equivalent) adhesion to sites of endothelial injury. Saw a few papers recently in CNS injury using transgenic lines to target 'pericytes' in the CNS & broad markers that are not specific, also label fibroblasts - imprecise tools & language = assigning functions in fibrosis to pericytes when other cells like fibroblasts are primarily involved 1/4

#KnowYourZDM: In 2019, a 10-year-old patient w/ a life-threatening lymphatic disorder was losing hope. Zebrafish to the rescue! Scientists created zebrafish with the same gene mutation and found a melanoma drug that fixed the fish. Then, doctors used the same drug on the patient to save his life! 🧪
Zebrafish are an excellent model for studying the development and function of blood and lymphatic vessels. Research in zebrafish has even led to treatments for lymphatic disorders in people. This image, taken using a powerful microscope that uses lasers to illuminate the fish, shows blood vessels (magenta) and lymphatic vessels (green) in the eye and head of an anesthetized, 6-week-old, transgenic zebrafish.

Many drugs have been repurposed for novel treatments such as COX inhibitors in leukemia & glucocorticoids in long QT syndrome. Figure taken from MacRae & Peterson (2015): How suitable are zebrafish for discovering human drugs? Although zebrafish enable in vivo studies on a large scale, questions remain about how relevant the findings are for drug discovery. Recent studies have begun to elucidate the degree of conservation between humans and zebrafish, which share 82% of disease-associated targets and a large number of drug metabolism pathways (as illustrated by the mass spectrometry of metabolic by-products). Zebrafish physiology is often well conserved (sometimes more so than rodent physiology); for example, cardiac electrophysiology (illustrative electrocardiogram (ECG) tracings from each organism are shown). Several compounds discovered in zebrafish screens have been shown to exhibit similar effects in rodent models and humans, including eight out of ten compounds tested by our groups. m/z, mass to charge ratio.
