Fascinating @science.org paper for chemsky from Ryan Shenvi’s group this week, where instead of making cyclopropane from a carbene and an olefin, they make it from three separate differentially reactive C1 sources in an iron-catalyzed sequence www.science.org/doi/10.1126/...

Iron-catalyzed [1+1+1] cyclopropanation by sequential coupling of an ester to two carbenes
Traditional precious metal catalysis benefits from well-characterized inner-sphere elementary steps in which a single complex binds and unites substrates at the metal center. By contrast, base metal c...

Think about how every single drug is different, and the focus in the discovery period is on how to make it work, and then they toss this ridiculous wild-looking molecule at the process team and say “Ok make a ton of it. Exactly the same way every time. With nothing dangerous!”

I love being honest with my medchem students when I say ‘I was discovery, that was easy- you only need 5mg.. but process? the art of making one molecule, perfectly, at manufacturing scale? it doesn’t get any harder or more beautiful than that.. now go read OPRD’

Plus they’re like you! They hate waste! It’s dangerous and expensive! And they’re making tons of material! Think about how they get that kind of precision while targeting safety and efficiency.

Just think first about the necessary precision and reproducibility constraints. Literal tons of material, and every single pill—every single one!—has to have the exact molecule at the exact dose and purity, or you don’t get better the way you’re supposed to.

If you follow me and you aren’t a chemist, just forget for a minute everything you think about pharma—for good or for ill—and briefly ponder how they made the drug you took last time you were sick. Not discovered its effects, but made it, at massive scale, under intense time pressure.

Biocatalysis is a growing part of organic chemistry, and I introduced it to my second-semester organic class several years ago. We teach the aldol reaction, we teach about enamines, we teach catalysis. Students who understand the aldol can understand an aldolase. (Those who don't… don't.) /2

Malfunctioning valve, that's a bad day
Firefighters first responded to the site in Garden Grove on Thursday, using water to cool the tank down as pressure began to grow inside it. But an attempt soon after by the company that owns the site, GKN Aerospace, to add a neutralizing agent to stabilize the chemicals was thwarted by the tank’s malfunctioning valves, Mr. Covey said.

That left emergency responders with no clear options to prevent the tank from failing.
