Glad to hear that it's worked well for you! The original Cth construct does work for on-bead cleavage, but in our hands, we ended up leaving ~3/4 of the protein we were trying to purify on the beads. Compare lanes 3 and 5 in this gel for on-bead cleavage using original flavor vs. new & improved

We can design "static" binders routinely now, but design of "dynamic" shape-changing proteins has remained quite hard. Jeffrey Chang and I asked: why can't we couple small-molecule binding to shape change? After all, natural proteins do it every day. In a new preprint, we show how it can be done. 🧵
