NAD depletion in skeletal muscle does not impair tissue integrity and function or accelerate aging, as shown in a mouse model with an 85% decrease in muscle NAD+ levels. Muscle structure, metabolism, and mitochondrial function remain unaffected, suggesting that NAD depletion does not drive age-related muscle decline.

Generation of self-organized neuromusculoskeletal tri-tissue organoids from human pluripotent stem cells
Modeling the neuromusculoskeletal (NMS) axis with organoids is challenging. Xiang and colleagues developed hPSCs-derived NNMS organoids (hNMSOs) with co-development, self-organization, and functional connection of neural, muscular, and skeletal tissues. hNMSOs revealed skeletal impacts on muscular development and neuromuscular function. These findings provide a model for studying human NMS crosstalk and disease.

Coren et al. show that in skeletal muscles, fibroblasts diversify into distinct subpopulations during embryonic development. These subpopulations reside in distinct niches, entail different dynamics and transcriptomes, and exert opposing effects on myogenic differentiation. This diversification process is dependent on muscle contraction in the embryo.

Gherardi et al. identify that aging and disability impair mitochondrial calcium uptake in skeletal muscle via MCUR1. Food-derived polyphenols increase the import of calcium in mitochondria by binding MICU1. The natural MCU activator oleuropein stimulates energy metabolism, ameliorates muscle performance, and reverses age-related muscle decline by increasing mitochondrial calcium.
