ROS are often viewed as damaging byproducts of metabolism but also play essential roles in cellular signaling. These findings suggest T cells need ROS to activate and that tumours exploit this dependency, releasing antioxidants into the tumour environment to suppress T cells

Cancer cells were the dominant source as CRISPR deletion of Prdx1 substantially reduced PRDX1 in TIF. Loss of Prdx1 drove T cell-dependent rejection of YUMM2.1 melanoma and sensitised otherwise refractory B16-F10 tumours to anti-PD-1/anti-CTLA-4.

Consistent with emergent roles of ROS in promoting signal transduction, antioxidant PRDX1 suppressed kinase signalling after just 10 mins of TCR stimulation (ERK, AKT, S6) while sparing proximal TCR events. SHP1/2 inhibition partially rescued signalling and effector function.

Remarkably, addition of the antioxidant protein PRDX1 to T cell cultures suppressed T cell activation and reduced intracellular ROS. Overexpression of NOX1 reversed this, indicating that PRDX1 suppresses T cells through depletion of extracellular ROS.

We set out to understand what makes the interstitial environment of tumours so suppressive to T cells, an interest that began ~15 years ago in Nick Restifo's lab with Robert Eil. Mass spec analyses showed marked enrichment of antioxidant proteins, with peroxiredoxin 1 (PRDX1) being highly enriched
