Also the SBF-SEM imaging captured microgamete entry into the macrogamete, revealing new insights into Plasmodium fertilization. This information is important for the development of potential transmission blocking vaccines to prevent malaria!

This revealed that the basal body of these cells has a unique arrangement and is unusually short, and that microgametocytes have a deuterosome-like structure that may provide a nucleating platform for the intiial basal body formation, allowing for the very rapid assembly of flagella

(1/4) We are very excited to share our latest preprint from @sauer-lab.bsky.social: Mega Expansion Microscopy (Mega-ExM)! We combine iterative hydrogel expansion with whole-proteome NHS labeling and post-expansion immunostaining, reaching tunable expansion factors up to ~1500× in whole cells 🤯

Importantly, knocking out the KIAP4 protein reduces Leishmania haptomonad adhesion and colonization of the sandfly stomodeal valve. This makes it these proteins an important target for reducing parasite transmission and therefore infection!

This work identifies the KIAP4 protein, a new component of the Leishmania adhesion complex, which is involved in parasite transmission. KIAP4 is part of a larger gene family that is found in other kinetoplastid parasite species like Trypanosoma congolense

The Sunter and Wheeler groups used Centre for BioImaging equipment to study the function of the ESB1 gene in the life-cycle of the pathogen Leishmania. ESB1 is located in a specific nuclear body, and it helps control expression of δ-amastins (cell surface glycoproteins important for infection)

There is also longer-range signalling between apical notches and regenerating regions. It appears that the central tissues can transmit this signal, but peripheral regions cannot. The phytohormone auxin is involved, with a balance between sources and sinks being important

It turns out that the apical notch has a substructure, that paracrine signalling appears to be important, and notches can reorientate in response to damage. Outside of the notches there is no clear predictability where meristems regenerate- apart from the oldest (stalk scar) cells not regenerating

The liverwort Marchantia makes clonal propagules called gemmae that are very accessible for experimentation and microscopy. This work explores the two (pre-)meristematic "apical notches" of the gemma and how meristem regeneration proceeds if these apical notches are removed or perturbated

Inflorescence meristem cell identities: (A) Force-directed graph layout of clusters associated with inner cell layers such as early primordia (EP), undifferentiated cells, procambium, xylem parenchyma and cortex.
