AMPA receptor gating proceeds through an intermediate pre-active state that has structurally remained an enigma. We used cryo-EM as well as full and partial agonists to resolve this state and for the first time structurally reveal the complete gating pathway of AMPARs. www.nature.com/articles/s41...

Voltage-clamp fluorometry turns 30 this year - thank you, Isacoff & Bezanilla! It has been a great tool to watch the outside of channels move, but of less use on the inside. That’s a shame because that is where a lot happens, incl. conformational changes related to sodium channel inactivation. 1/x

Localized covalent targeting of NMDA receptors reprograms behavior
Spatiotemporal manipulation of neural circuits relies on genetic tools which require recombinant expression of exogenous actuators. Bátora et al. show that localized covalent inhibition of endogenous NMDA receptors is sufficient to reprogram behavior. Their two-photon molecular tattooing approach enables persistent, region-specific modulation of neural circuits without genetic manipulation.
