Heterologous Rieske Nonheme Iron Monooxygenases Enable Efficient Microbial Conversion of Lignin Guaiacol to Adipic Acid
Adipic acid (1,6-hexanedioic acid) is a key building block for nylon-6,6, a widely used polymer in the global chemical industry. Current industrial production relies on petrochemical feedstocks and nitric acid oxidation of cyclohexane/cyclohexanol mixtures, releasing nitrous oxide, a potent greenhouse gas. Biotechnological routes offer sustainable alternatives but have been limited by low yields or reliance on multistrain systems. Here we report a one-pot, single-strain microbial process for the efficient conversion of guaiacol─a lignin-derived aromatic─into adipic acid. By integrating heterologous Rieske nonheme iron monooxygenases from Cupriavidus necator N-1 with systematic process optimizations in engineered Escherichia coli, we achieve near-quantitative conversion with 97% yield and titers of 1.5 g/L in aqueous, lab-scale reactions. This work demonstrates a novel and efficient strategy for lignin valorization through engineered microbial synthesis, providing a new sustainable and scalable route to adipic acid.

It's hard to describe how valuable CASP is to the structural biology & structure prediction field. The ever increasing number of prediction algorithms means independent assessment of their accuracy is vital. Please contribute interesting/tricky structural data if you can!

Recycling rates for PET are shockingly low. The more we show the value in collecting & treating "waste" PET, the more we encourage wholehearted participation. If Incentives aren't strong enough to recycle PET for rPET at $2 per kg, what about if your product is $50K per kg? What else can we make?

For me this is a really important piece of work- beyond the fundamental understandings applicable to other researchers (use of transporters, identification of intermediate inhibition, product degradation route) it highlights the potential for PET plastic as a valuable precursor.

There's a lot of discussion whether economic growth & carbon emissions can be decoupled. Some critics say countries (e.g UK) reduce emissions while growing by offshoring pollution (i.e. to China). This is a fantastic rebuttal- China continues to grow economically with reduced CO2 emissions.

If this is true, it’s abysmally shortsighted for #NIH. The new tools nurtured by the long series of #CASP challenges probably contribute far more in one year to the scientific and industrial economies than the entire funds that have supported it.
