First paper entirely from our lab is out in the wild! We tested if reprogramming still works in the aged CNS and found the aged retina blocks neurogenesis both intrinsically in Müller glia and through exacerbated inflammation. Then point at some ways to overcome this. www.pnas.org/doi/10.1073/...

Honored to receive the Postdoc Winston Fellow Competition Award from the Department of Ophthalmology at WashU. This award followed my presentation on the requirement of DNA demethylation for retinal development, cell‑fate specification, differentiation, and function. The best is yet to come!

In the spirit 👻 of Freaky Friday 🎃, here is a beautiful yet eery video of zebrafish retina development showing horizontal cells (🔵) finding their way out of the crowded amacrine (🟠) layer to settle beneath the photoreceptors. Happy #Halloween! 🎥: PhD student Rae Wong from the @nordenlab.bsky.social

For today’s #FluorescenceFriday, the #rods clearly chose chaos What you’re looking at are long, beautifully coiled outer segments of rod #photoreceptors… basically the retina showing off its inner spaghetti aesthetic. Elegant, tangled, and absolutely essential for seeing in low light.

Looking through old photos and found this beauty. Frog rods, labelled with GFP under a XOP promoter (inner segment, orange) and wheat germ agglutinin (outer segments, cyan). I love how you can see the holes where the mitochondria are packed near the top of the inner segment (the ellipsoid zone). 🧪🐸
Frog photoreceptors showing a blue outer segment and a bright orange inner segment. Near the top of the orange inner segment is an area with small holes, where mitochondria are packed in very tightly to support the metabolic needs of outer segment synthesis. This area is called the ellipsoid.

Excited to share our new manuscript from the Yoshimatsu and MacDonald labs. www.biorxiv.org/content/10.6.... We found that my favourite glial cells can regulate local retinoic acid signalling to specialise cone photoreceptors for high acuity visual function.

#FluorescenceFriday The #seahorse #retina shows off its beauty — stunning stratification in the inner nuclear and inner #plexiform layers, with rod #bipolar terminals forming a brilliant band. #Vertebrate retinas: the same basic plan, yet each species adds its own twist. Its so fascinating.

#FluorescenceFriday #Seahorse retina stained with #GNB3 (green) + #DAPI (cyan). GNB3 labels cone photoreceptors + ON #bipolarcells, tracing pathways from outer to inner retina. Amazing to see conserved visual #circuits in this non-traditional model

Happy to share the definitive version of our last paper about the role of TET enzymes and DNA demethylation in retinal development @clark-lab-retina.bsky.social journals.plos.org/plosbiology/...
journals.plos.org
Active DNA demethylation upstream of rod-photoreceptor fate determination is required for retinal development
TET enzymes remove DNA methylation markers, but the role of this process in retinal development is not clear. This study shows that these enzymes are required for photoreceptor cells to initiate the g...

We recently wrote a review about neuroimmune interactions in retinal repair. We dive into the literature regarding immune influences in Muller glia reprogramming, cell transplantation, and axon regeneration. Hopefully its useful, free link: authors.elsevier.com/a/1k-FL4pAsj...
