Through additional analyses (metabolite effects, GRS PheWAS, MR) we drill down on the discordant risk mechanisms, identifying liver fat accumulation as a nexus of discordant risk. But not all liver fat is equal, different mechanisms of hepatic fat accumulation come with different disease risks.

To try and understand these patterns we took a variant clustering approach (bNMF) which has been useful for partitioning T2D-risk pathways using GWAS variants. But here, we cluster shared variants, and identify finer-scale clusters than what has previously been observed for T2D.

Excited about a new preprint from the lab looking at pleiotropic variant effects between type 2 diabetes and coronary artery disease. In our analysis we identify 42 unreported shared loci, but most of them (35) have discordant effects on these co-occurring diseases! www.medrxiv.org/content/10.6...

Was fun being back in Toronto last week to celebrate the 20th Anniversary of the @donnellycentre.bsky.social (thedonnellycentre.utoronto.ca/news/looking...). Cool opportunity to present and think about my academic journey and switching fields since my PhD! But best was catching up w/ old colleagues!

@omicspred.bsky.social centralises imputation models from multiple resources (PredictDB, Zenodo, INTERVAL/UKB-trained scores) and multiple omics layers (transcriptomics, metabolomics, proteomics) to make analyses easier and more comprehensive, e.x. this multi-ancestry multi-omics MVP PheWAS!

Interested in applying multi-omics imputation or TWAS/PWAS approaches to your data? Frustrated by the models being spread across multiple resources? Enter OmicsPred: a centralised resource for genetic prediction of multi-omic traits! 🔗 : www.omicspred.org 📃 : www.medrxiv.org/content/10.6...

Clinical statements (AHA, ESC) have suggested that PRS may be most useful as a risk-enhancing factor for those at intermediate risk. We used PRS to reclassify individuals at intermediate risk by QRISK3 and showed improved sensitivity and fairness benefits (particularly for age & sex).

We evaluated PRS in two contexts, as a standalone risk predictor and as a risk-enhancing factor. Given the known performance differences across ancestries, we were surprised to find that the fairness of PRS predictions across ethnicities was equal to or lower other CVD risk factors when used alone.

Interested in methods for updating risk prediction models (time-to-event) with new features? Check out this new paper with Will Underwood & co introducing the CARE method, and an application where we augment SCORE2 with PGS & IMD: arxiv.org/abs/2506.23870.

Great fun sharing the @pgscatalog.bsky.social at the pre- #ESHG25 course on open-source genomics resources. Thanks to Juliana Cerqueira and Inga Prokopenko for the invite! Looking forward to the rest of the conference!

Congratulations @clairecoffey.bsky.social on passing your PhD viva (it's not everyday your first PhD student graduates)! It's been a pleasure to supervise your work applying algorithmic fairness techniques to risk prediction models and polygenic scores with Angela Wood & @mikeinouye.bsky.social.
