As is tradition at Georg-August-University Göttingen, Gesine had to climb up to the Gänseliesel to give her flowers. Less traditionally, but very much in the spirit of the Huisken lab, Jan gave Gesine a ride to the Gänseliesel on a cargo bike. @uni-goettingen.de #Gänseliesel #Göttingen

We fluorescently labeled embryos and recorded whole-embryo time-lapse movies with our portable Flamingo light-sheet microscope. We’ll reconstruct the cell lineages and compare wild-caught embryos with our lab strain. Do annelid embryos develop differently in the wild? We are excited to find out!

Platynereis dumerilii uses the lunar cycle to synchronize sexual maturation. At night, the worms swim to the surface to reproduce in a spectacular nuptial dance. We collected mature worms in Villefranche-sur-Mer and set up matings in the lab to obtain fertilized eggs for our experiments.

Do wild-caught embryos develop in the same way as embryos cultured in the lab? This summer, Beke and Mette visited Luis at IMEV in Villefranche-sur-Mer, France, to start exploring this question in the marine annelid Platynereis dumerilii. #Platynereis #annelid #Flamingo #lightsheet #cell-lineage

#FluorescenceFriday ⚔️ DNA-PAINT vs self-blinking dSTORM, with #Vimentin as the benchmark structure. PAINT: tunable, multiplexable, almost no bleaching. SB-dSTORM: fast, buffer-independent, user-friendly, ExM-friendly. So who takes the belt? 🤼 The fight is on. 😄

Isotropic, aberration-corrected light sheet microscopy for rapid high-resolution imaging of cleared tissue - Nature Biotechnology
A light-sheet microscope built from off-the-shelf components enables high-resolution clear tissue imaging.

The same microscope can image larger samples, including a whole mouse brain (1 cm^3), and supports hybrid low- and high-resolution modes. Low-resolution imaging gives a full overview, while high-resolution mode enables multi-ROI isotropic submicron imaging on the same specimen.
011
Huisken lab @huiskenlab.bsky.social · 14/11/2025
The system also supports larger samples at higher RI (e.g., 1.56), such as mouse cochlea, which are dipped directly into immersion media to image neurites and reconstruct individual connections between spiral ganglion neurons and inner hair cells across the entire cochlea.
110
Huisken lab @huiskenlab.bsky.social · 14/11/2025
The microscope can image fragile samples like cleared zebrafish, RI-matched to 1.47 and held in a glass capillary, allowing isotropic sub-micron-resolution imaging of the vasculature.
110
Huisken lab @huiskenlab.bsky.social · 14/11/2025
Together, these optical innovations enable us to cover a wide range of biological samples, including entire zebrafish vasculature and mouse organs such as the cochlea and brain vasculature.

Huisken lab @huiskenlab.bsky.social · 14/11/2025
We achieved isotropic sub-micron resolution with an axially sweeping light sheet running near the camera’s top frame rate. A high-frequency closed-loop system stabilizes the remote-focusing voice-coil motion, enabling 100 fps while preserving sub-micron resolution over a large FOV.
110
Huisken lab @huiskenlab.bsky.social · 14/11/2025
Unlike other light-sheet systems that use the remote-focusing arm only to sweep the sheet, we found it can also correct distortions such as field curvature. By using a concave instead of a flat mirror, we compensate curvature and expand the usable FOV across the full RI range.
110
Huisken lab @huiskenlab.bsky.social · 14/11/2025
Our microscope uses an air objective lens for illumination. To make the air objective suitable for creating a diffraction-limited light sheet in high-RI media, we use an off-the-shelf meniscus lens with a surface curvature matched to the NA of the illumination lens across all refractive indices.
110
Huisken lab @huiskenlab.bsky.social · 14/11/2025
Our compact, high-speed light-sheet fluorescence microscope is built from off-the-shelf optics to achieve 850 nm isotropic resolution for cleared samples up to 1 cm3, across refractive indices from 1.33 to 1.56.
