Today I'm posting a new blog! Nearly two hundred papers have muddled the names for different proteins and recorded using the wrong antibody for a range of experiments. This time it's PERK vs. p-ERK. Including researchers from Harvard, the Mayo Clinic, and more. sholto.substack.com/p/perk-vs-p-...

Congratulations @prithadg.bsky.social ! This was a fun project that allowed me to mess about on computers for a little bit, learning how to design de novo protein binders.

If it helps, I've had a pair of goodrs for a couple of years now and only had the one near-miss with a housecat. Assuming that holds true for bigger cats, they're probably about 92.6% leopard-proof.

In accordance with ancient tradition, our next Prime Minister is announced to the nation while standing between a furry and a man with a bin on his head.
A victorious Andy Burnham MP (24,937 votes), as the result of the Makerfield by-election is announced, flanked by defeated candidates Count Binface (a bloke in a vaguely sci-fi costume with a large bin for a helmet; 95 votes) and Robert Pownall (who is in a full body fox costume, holding a sign reading "Protect British Wildlife"; 18 votes). There are some other candidates there just out of shot but that's not important right now.

🚨⚠️ New Preprint Alert! ⚠️ 🚨 www.biorxiv.org/content/10.6... Screening p97 cofactors, @prithadg.bsky.social identified FAF2 as the strongest activator of the p97-UFD1-NPL4 complex. Leveraging her mechanistic descoveries, we designed 𝘥𝘦 𝘯𝘰𝘷𝘰 binders that could also enhance p97 substrate unfolding.
Image of FAF2's predicted helical domain interacting with the UFD1 UT3 domain. Next to this are similar images showing the predicted binding of de novo designed protein binders to the same region. Above is a sequence alignment indicating the key residues involved in the interaction.
