The logic is strikingly reminiscent of the Cdk–cyclin system: partially overlapping, stage-specific activators dictating when and where a single kinase acts.

Cep192, together with Aurora-A, drives Plk1 activity in S phase, where it controls replication origins firing. Bora takes over in G2 as dominant activator, driving mitotic entry, Checkpoint recovery, and centrosome maturation. Centrosomal activators cannot substitute for it, despite sitting there.

The reason this was easy to miss comes down to how Plk1 is switched on. It has three independent co-activators, cytoplasmic Bora, and the centrosomal proteins Cep192 and Cenexin. The obvious assumption is that they provide redundancy, however, they don't. Each one activates Plk1 for a distinct job.

Our new paper is out @natcomms.nature.com Plk1 is one of the master regulators of cell division. We found a low level of Plk1 activity is already present in S phase, and this activity sets the pace of DNA replication. www.nature.com/articles/s41... #CellBiology #Mitosis #Centrosome #OpenAccess
