Is euchromatin really “open”? 🧬 Using super-resolution imaging🔬 our new study @natgenet.nature.com reveals: Euchromatin forms condensed domains in live cells. Cohesin constrains them and prevents domain mixing for proper transcriptional insulation🚧 🔗 www.nature.com/articles/s41... (1/2)

😊 New preprint! In budding yeast, Dna2-intrinsic condensation regulates long-range DNA-end resection. Analyses suggest redistribution of condensate grammar in humans. Congrats to Chris Jordan and the Mekhail, Wyatt, Cowen & Hakem labs! More to come soon. www.biorxiv.org/content/10.6...

Dna2-intrinsic condensation regulates DNA end resection and reveals evolutionary redistribution of condensate grammar
Eukaryotic cells commonly use biomolecular condensation of DNA double-strand break (DSB) repair scaffolds and signaling assemblies to organize repair reactions in space and time. Yet whether DSB end-p...

Thrilled to share a new important study from the lab, published today in @nature.com, showing how cohesin mediates efficient replication fork plasticity and stress response, using its loop extrusion activity to promote 3D contacts on replicating DNA: www.nature.com/articles/s41...

Nested TAD hierarchy defines cohesion zones on replicated chromosomes - The EMBO Journal
During each cell cycle, cells must not only duplicate the DNA sequence but also preserve the three-dimensional genome architecture required for gene regulation and DNA repair. After replication, this ...

RNA reinforces condensate nucleation on chromatin to amplify oncogenic transcription
Budinich et al. show that locally produced transcripts reinforce the nucleation of pathogenic condensates formed by mutant ENL at permissive target loci. This RNA-enhanced condensate nucleation in turn increases mutant ENL occupancy and transcriptional bursting at key oncogenic targets, thus surpassing the gene activation threshold required for tumorigenesis.
