💯. I think too many people treat AI containment and alignment as an engineering challenge to be solved. As I see it, the problem is far more fundamental, and harder to solve (arguably unsolvable). Understanding why requires understanding evolution. Short thread 🧵

result underscores the importance of developing and understanding effective combination therapies. My UNC colleague Adam Palmer is doing some excellent work investigating this topic. You can find him on the other place: x.com/ac_palmer (5/5)

We found that sometimes sequential therapy outperforms combination therapy. This is because sequential therapy can sometimes purge mutations that provide a lesser degree of resistance to both drugs. But the param space where this is true is smaller than in our original submission (bug fix). This...

remove as much standing variation as possible so that a second-line treatment has better odds of driving the cancer cell population to extinction. The timing of the switch from the first to second treatment matters a lot here, and we are investigating this in a follow-up study Another update: (3/5)

The biggest change from the preprint version is that we found a bunch more evidence that, perhaps unsurprisingly, standing variation plays a super important role in the evolution of treatment resistance. The goal of a good treatment is thus twofold: 1) kill cancer cells, and 2)... (2/5)

In small pops, the initial stochastic phase (conditioning on the allele not being lost) isn't much slower than for codominant/dominant alleles, both of which (especially dominants) are slower than recessives toward the end of the sweep. Hence, recessives don't have more opportunity for conversion.

during revision. For example gene conversion is more likely to "soften" sweeps of recessive beneficial alleles (which have a longer sojourn than codominant or dominant alleles and thus more opportunities for gene conversion), except for in small populations.

Obviously it took me an embarrassingly long time to get around to the revisions here. I had been trying to prioritize trainees' projects and this thing got left on the backburner for way too long. But I enjoyed finally getting back to it and I think there are some interesting tidbits that were added

If they keep using MYF for this fraction of new awards, and end up spending all of their budget for the fiscal year, would this end up running afoul of the stipulation in the appropriations bill that MYF be capped at the same level as last year? Or am I misunderstanding something here?

Can we predict what the overall success rate for new grants should be this year based on this rule? Wondering if this is roughly the same as last year or if other shenanigans going on last year muddy that a bit. (Not that there won't be new shenanigans this year, but happy to ignore that for now...)
