Discovery of CDK4-selective molecular glue degraders by high-throughput proteomics
Molecular glue degraders (MGDs) are proximity-inducing molecules that promote the destruction of disease-causing proteins by stabilizing novel interfaces between E3 ubiquitin ligases and target protei...

As part of a prolific collaboration with the ICR Centre for Protein Degradation, we just recently described a glue degrader engaging the homodimeric interface of TBK1. doi.org/10.64898/202... Curious to see how our understanding of CRBN neosubstrate recognition will continue to develop...

Developing potent and selective TBK1 molecular glue degraders for cancer immunotherapy
Immune checkpoint blockade (ICB) has transformed cancer therapy across multiple tumour types, yet primary and acquired resistance remain major barriers to durable benefit. TANK-binding kinase 1 (TBK1)...

As the list of CRBN neosubstrates grows longer (doi.org/10.64898/202...), we and others find more and more glue targets with non-canonical binding modes.

Integrated proteomic screening reveals design principles of CRBN molecular glue degraders
Cereblon (CRBN)-based molecular glue degraders (MGDs) induce the degradation of diverse disease-relevant proteins, underscoring their broad therapeutic potential. Here we systematically expand the CRB...

Pivoting the proteomics platform from high-throughput screening to validation, SAR, and mechanistic exploration, we not only optimized the degradation characteristics but also uncovered our glues to recruit CRBN specifically to CDK4:INK4 heterodimers.

Excited to share my first major internal research project at NEOsphere! Generating global proteomic profiles for entire cereblon glue libraries, we identified a degrader of the master cell cycle regulator CDK4.

This project started 5 years ago. It led us to add isotope-labeling support to #FragPipe/#IonQuant. Since then, the tools have grown so much and are now widely used in #Chemoproteomics. Huge thanks to everyone, and special thanks to @stephanhacker2.bsky.social and @pzanon.bsky.social

I am thrilled to see the paper out now and grateful to all coauthors for their contributions to this work! Among a number of changes, we included additional probes as well as experiments in live bacterial cells. So be sure to check it out even if you are already familiar with the preprint!

Some cuts have been starkly visible, but the country’s medical grant-making machinery has also radically transformed outside the public eye.
