@noahnturner.bsky.social, Barford et al reveal conserved underlying mechanisms of complex assembly, auto-inhibition, & phosphoregulation that ensure precise control of KMN assembly onto #centromeres to enable genome division. rupress.org/jcb/article/... #kinetochores #cryoEM #biochemistry #genetics

This long, challenging project would not have been possible without the insight and experimental work of my co-authors Ziguo Zhang, Jing Yang, Kyle Muir, Stephen McLaughlin (@lmbbiophysics.bsky.social), Tomos Morgan and my PhD supervisor, David Barford. Thanks to everyone involved!

Our work reveals conserved principles that could not be previously discerned by which the KMN complex assembles and functions in humans and yeast.

We found many interfaces that hold the budding yeast complex together and regulate its assembly onto chromosomes have diverged from those that serve these functions in humans. Despite this, the architecture and functional principles of the human and budding yeast complexes are strikingly similar.

We therefore used biochemistry, biophysics and AlphaFold predictions to further clarify the molecular mechanisms by which the budding yeast KMN complex functions.

To investigate this fascinating and complex machine, we analysed the structure of reconstituted budding yeast KMN complexes using cryo-EM. Our cryo-EM density maps were limited to a maximum resolution of 4.2 Å, which we could only obtain by utilising Blush regularization implemented in RELION 5.0.

When assembled onto DNA, the eukaryote-specific kinetochore grabs on to shrinking microtubules to power chromosome segregation during mitosis. When this process goes awry, cells inherit an incomplete copy of the genome and can become sick or die.

Happy to see our paper describing cryo-EM structures of the budding yeast KMN kinetochore complex published in @jcb.org! Our structure reveals how an important hub mediates and regulates assembly of the machine that segregates chromosomes. #cryo-EM @mrclmb.ac.uk 🔗 rupress.org/jcb/article/...
