more speculation than substance here. while it makes sense to take reasonable precautions w/ unknowns, i'm just not convinced LLMs are anything like the major bioweapons catalyst it's being hyped as. (plus you can't ignore that this is good PR for anthropic. "see! we can regulate ourselves" etc.)

I see lots of hand wringing and hot takes on why an mRNA vaccine worked in melanoma and not CRC. The hot/cold tumor aspect isnt even the main part of the discussion to me. We know IFNg upregulates PDL1, causing vaccine failure. Why would BioNTech even bother with a monotherapy arm?

tl,dr; Scientists no longer allowed to use federal funding to publish, attend meetings, or talk to the public. They cannot collaborate internationally. Grants can be cancelled for any reason, at any time, political appointees have a final say over what gets funded, and who gets funded.

So let me say it as clearly as I can: Without basic scientific research, supported by the kind of farsighted public investment that allows large-scale, undirected, curiosity-driven inquiry, the scientific pipeline will run dry.
In daily life, people may not feel the effects right away, or even in 10 years. But we will feel it. And when someone we love needs therapies that could have emerged but didn't or when other countries now
investing in science can launch new science-based industries or run their societies on vast resources of fusion energy or reap the benefits of quantum computing power or advanced medical breakthroughs, America will wish it sustained its leadership in scientific research here and now.

Our rapid enrollment and results demonstrate that patients want, and there is scientific rationale, for sequencing immunotherapy first without a "kitchen sink" approach of combining it with chemotherapy. To quote one of our patients "I wanted a chance to avoid chemo, even if that isn't forever" 6/n

While this data is still maturing, BBOpCo shows what is possible in a disease like MSS CRC that has no approved immunotherapies by applying an anatomically informed patient selection approach to expand treatment options. 5/n

Median freedom from crossover to chemo was 8.7 months, meaning BOT+BAL delayed chemo, and our DCR after crossover in 4 evaluable patients was 75%, indicating that our immunotherapy first followed by chemo rescue strategy is feasible. 4/n

With a median follow up of 5.4 months, we observed a 75% disease control rate (DCR) and already have 2 confirmed iPRs to BOT+BAL alone, one in a patient with lung mets and another with peritoneal metastasis, with tissue TMB of 1 and 3 mut/mb respectively. 3/n
