At the #ACSFall2026 drug candidate first disclosures session yesterday, several molecules' debuts were also their swan songs. One of them caught my eye when I looked at abstracts: an inhibitor of lipogenesis (that's synthesis of new fatty acids) aimed at treating acne. 1/

SGLT2 inhibitors activate pantothenate kinase in the human heart
Sodium-glucose cotransporter 2 inhibitors (SGLT2i) reduce mortality in heart failure, but their pharmacological target remains unclear. In this study, we showed that SGLT2i directly activate pantothen...

It will also be a potential benefit to a few rare disease communities (there are a few in born errors of metabolism where a PANK activator would make sense). A few specific med chem groups have been working on it for a while, excited to see full industrial shops get interested.

It was so much fun because getting into this project I thought it would be ketone related from prior work. Finding out it was CoA related is just a absolutely *chefs kiss*

I think the job interviews there must be fun because they must need a few people who are subtly bad in lab and can’t figure out why.

Thrilled to see our work on senescence & metastasis out! We found that chemotherapy increases ovarian cancer metastasis in a paracrine manner via fructose. Dietary fructose is sufficient to increase metastatis. This work has multiple implications related to both chemo and diet.

And please put all of this and more in your comments on the RFI. It’s also notable the co-I subcontracts are all of the pain of MPIs but much less stable and equitable to the collaborator as they could be easily cut, could not be used for supplements, and didn’t show up in center or T grants.

It's time to rewrite reviews-aKG doesn't just regulate demethylation. Work from my lab and @mzspectrum.bsky.social demonstrates a new role for aKG in promoting histone acetylation and DNA repair through regulating carnitine synthesis. Congrats Apoorva Uboveja! www.nature.com/articles/s41...

The lab is expanding and hiring both a postdoc and staff scientist! See more on our website: airdlab.com. We study how metabolism and cell cycle states drive tumor progression, therapy response, and immune interactions. Candidates interested in mechanistic cancer biology are encouraged to apply.

Notice says PI/MPI. So not co-I. even if your institution says you are PI of subaward NIH doesn’t count that. Being said- subawards have triggered well funded investigator review issues (ask me how I know). UC Davis site has good plain language explanation.

Anybody got an example of substrate channeling where an isotopic label in a specific product is more enriched than the tracer? I know this is theoretically possible with compartmentalized pools but I am drawing a big old blank on relevant literature?

I read that too and wondered how much was CR funding based. I’ve had budgets cut and the PO allowed reduction in scope.

Found out my well scored MIRA will not be funded, as I am MPI on too many grants. Not close to the direct cost cap at all, but the optics of 5 projects look bad. I understand that and really hope the grant ends up keeping someone else’s lab open. But man it hurts.

Happy New Year. I’m pleased to report publication of the Ducker lab’s latest cancer metabolism paper led by graduate student Kyle Dunlap in @cp-cellreports.bsky.social www.cell.com/cell-reports.... 1/n

SLC7A5 is required for cancer cell growth under arginine-limited conditions
Metabolic adaptations are key to cancer cell survival in nutrient-limited microenvironments, but strategies for coping with arginine scarcity are poorly understood. Dunlap et al. use CRISPR screening and metabolomics to identify the plasma membrane transporter SLC7A5 as a citrulline transporter essential for cancer cell growth under arginine limitation.
