Glad to see this work finally out! A joy working with the incredible Nurun Fancy to figure out the interaction between TREM2 and CD33, how TREM2 variants differ and what happens to other cell types in cases with TREM2 variants. Thanks to everyone involved in the study, it was a great team effort!

Pleasure to collaborate with Jo Anne Stratton @theneuro.bsky.social funded by @hbhlmcgill.bsky.social and @ukdri.ac.uk. We show that post mortem processing conditions have differential effects on the brain's transcriptome. And temperature may play more of a role in good quality brain tissue.

Taken together, we believe we have derived a pseudotemporal profile of synaptic changes in human AD and shown that they are associated with changes in the astrocyte proteome, particularly that of PAPs. 5/6

Alongside these synaptic changes, we found a astrcoyte signature that was enriched for proteins at the peri-synaptic astrocytic processes (PAPs). This astrocyte signature increased with pathology and was associated with reduced synaptic resilience. 4/6

Mitochondrial proteomic changes dominated in the low pathology region, inhibitory changes in the mid pathology region and widespread synaptic transmission pathways were most prominant in the higher pathology region. 3/6
