Well said on the role of somatic mutations in myeloid cells in Alzheimer's disease! (www.cell.com/trends/molec... The CSF2RB A455D variant from our study is also a promising example. More studies using chimeric brain models with human microglia carrying somatic mutations are needed.

ApoE3 R136S binds to Tau and blocks its propagation, suppressing neurodegeneration in mice with Alzheimer’s disease
Guiqin Chen, Mengmeng Wang, et al. demonstrate that ApoE3 R136S inhibits AEP-mediated Tau cleavage and neuroinflammation by binding more tightly to Tau, preventing its propagation, and thereby slowing the progression of Alzheimer’s disease in mice.
