2/ ProPER implements in situ proximity extension by leveraging primer exchange reaction. The hairpin extends its cognate primer when they are in physical proximity. Proximity-encoded DNA barcodes are recorded into single stranded DNA concatemers, which are read out by fluorescent oligos @embl.org

1/ ProPER - Our novel ligation-free proximity detection method for efficient multiplexed imaging of molecular interactions in situ. Among other things we use it to image the life cycle of RNA molecules and resolve differential kinetics of gene expression steps. New preprint: doi.org/10.64898/202...

Ever felt constrained by the "4-color limit" in fluorescence imaging? We use DNA-barcoded labeling to build high-plex panels, enabling imaging of 15 targets across the full fluorescence spectrum. doi.org/10.64898/202... Grateful to @sinemsaka.bsky.social and all authors for making this possible.

Whole-brain connectomes are rare and expensive “hero datasets.” 🧠 Dr. Sven Truckenbrodt explains why cost and scalability limit comparative #connectomics, and why that matters for #neuroscience. Check out the full episode here: youtu.be/a4qpFpjnOX4
