One especially cool thing was Sangyoon’s use of machine learning-based analysis of ultrasonic vocalizations (based on this paper: pubmed.ncbi.nlm.nih.gov/28472651/). Exon 3 mice exhibited an impoverished vocal repertoire, reminiscent of the communication deficits observed in ASD.

This distinction mattered. Ptchd1 exon 3 mice exhibited a broader autism-relevant phenotype, including social and communication deficits not observed in the exon 2 model (pubmed.ncbi.nlm.nih.gov/27007844/)

We found that deleting exon 2 does not necessarily abolish PTCHD1 function because it triggers compensatory upregulation of the Ptchd1-C isoform. In contrast, targeting exon 3 disrupts both major brain isoforms (Ptchd1-A and Ptchd1-C), producing a more complete loss-of-function model.

Human genetics provided the motivation. Pathogenic PTCHD1 mutations in patients are concentrated in exon 3, yet previous mouse models primarily targeted exon 2.

New paper from the lab, led by Sangyoon Ko (and a long-term collaboration with John Vincent at CAMH). The study revisits a basic question in modeling PTCHD1-associated autism: which exon should be targeted to best capture the human disorder? 🧵 can01.safelinks.protection.outlook.com?url=https%3A...

But overall… a bit disappointing. Crust looks great, but bread too dense for my taste. Not great, not terrible. But cafe worth a visit.

New baguette alert. Been wanting to try Liberty Village’s Brodflour for a while, and today I got the chance. First of all, extra points for cool cafe vibe. Busy inside, and lots of people gathered outside with coffee cups doubling as hand-warmers.

I'm always amazed by the complexity and nuance of the neuroscience of pain. In this Perspective, @simonbeggslab.bsky.social and @franklandlab.bsky.social do a great job of explaining some important new findings on the role of time in the mechanisms of chronic pain www.science.org/doi/10.1126/...

Save the date for the annual "Neural Circuits and Behaviour" satellite conference at the Canadian Association for Neuroscience annual meeting, cohosted by @franklandlab.bsky.social and myself on May 18, 2026. We have a great speaker lineup and will be selecting 4 trainees talks from our registrants!

This baguette is spot on. Airy, with near perfect crust. Delicious with some Wensleydale and some Branston! It’s up there with the best baguettes in the city (Bricolage and Amadeus). Need to do a side-by-side comparison.

These time-dependent shifts in sub-engram engagement indicate that systems consolidation involves reorganization _within_ (as well as across) brain regions.

We provide evidence for projection-defined 'sub-engrams' that contribute differentially to memory expression at recent and remote delays.

This adds to an emerging picture that not only do new neurons influence encoding of new memories but they _shape_ and _remodel_ existing memories. These shifts in resolution may facilitate generalization of event based memories beyond the original situation.
