And even if it cannot be excluded (and in modularity and diversity is the beauty of EXO70s), when there is evidence of EXO70.2 subfamily members cooperating with the complex, I would probably not buy it as it`s ancestral state.

Agreed, it generates robustness which you can trust and build on. Hard to say without mutants, but MpEXO70.2 OE ripped epidermis phenotype resembles OE of FER/MRIR240C doi.org/10.1242/dev.... and ropgap/ren doi.org/10.1242/dev...., so maybe something with cell wall integrity/recycling?

Thanks! Yes, we made an independent attempt to interpret plant exocyst evolutionary dynamics (also by using DNA from algae!) I would say both papers are worth reading, maybe even side-by-side #PlantScience

Thank you, I am glad you liked it! It`s never easy to find oneself (even if only partially) being "scooped":D Have been working on it since 2019, but after not being able to get KO mutants in MpEXO70s, I decided try perhaps slightly more avant-garde approach presented in preprint

Altrough I do agree, couldn't it be in the end just pragmatical? For publishing in "non-profit" society journals you pay circa the same price (without transparency where the money goes to) for lower IF (important for your further evaluations)?

Have been keeping whole sporangias in a freezer on -20 without notable germination problems so far. Possibly two issues: spores were not matured (you harvested it too soon), or they were not properly dehydrated (air-dry them after collecting and/or add a few silica gel beads for some time).

Thank you, this looks like a lot of work! If I may ask, why you named it "Evolution" and not "Morphogenesis"? Based on the title, I was expecting slightly different content - not too much is known about the evolution of phyllids (and the preprint is not touching it also)

What works for me best in those cases: go to the primers they used, blast it against genome/transcriptome of the plant - uala, you have got the protein

(1/6) First and foremost, Auxenochlorella spp. are oily green algae that can be readily transformed by homologous recombination, enabling efficient site-specific transformation of the nuclear genome. This is a first for chlorophytes, and we hope that it will facilitate lots of interesting research

Yes, that's right. Just wanted to try to pinpoint sometimes quite unnecessary workload accompanying the neutral locus game (in this case you would need to perform ca 10x more transformations to proove the same point). Anyway, thank you for the insights!

So able to get it to the plant, it's working, but not enough to pick ones integrated correctly. Now you are in the situation, where you are able to show things, but becouse neutral locus fetish (I think that it wouldn't be much different if they would be integrated correctly), it's unpublishable

This is fine and I agree with that. But for example, now I do have a double mutant with a severe phenotype and wanted to put things to the neutral locus. The mutant transformation efficiency is super low (ca 3 transformants per dish).

But that's actually the thing, no? In randomly integrated KI lines, you will pick few of them which are performing the best. And they are proving your point, becouse of the randomness (and still are most probably in undefined 'neutral locuses' becouse there will be thousands of them in the genome)

Yes! This is what I am successfuly emplying now, I call it knock-knock system, since at first you knockout gene by HR from start to stop, and then you just replace the resistance in plasmid, insert the CDS and resistance and use it for knockin. Works great for single copy genes
