Sialic Acid-Containing Glycolipids Extend the Receptor Repertoire of Enterovirus-D68
Enterovirus D68 (EV-D68) emerged as a pathogen of increasing health concern globally, particularly due to its association with outbreaks of severe respiratory diseases and acute flaccid myelitis (AFM) in children. Knowledge regarding the tissue tropism and pathogenesis of EV-D68 within the respiratory tract and central nervous system remains limited, primarily due to an incomplete understanding of the host factors that facilitate the entry of EV-D68 into host cells. Several cellular receptors involved in EV-D68 infections have been identified, including ICAM-5, sialylated glycoproteins, and heparan sulfate (HS). Here, we investigate the receptor requirement of a panel of EV-D68 strains covering all clades, focusing on HS and sialosides utilizing glycan arrays. We found that all EV-D68 strains binding to HS harbor a cell culture adaptive substitution in the structural protein VP1 at position 271, which changes the amino acid into a positively charged one. Glycan array analyses revealed that EV-D68 strains prefer α2,6-linked sialic acids presented on N-glycans, α2,8-linked sialic acids on gangliosides, or both. Inhibition of glycolipid biosynthesis or multivalent glycolipid mimics confirmed that ganglioside structures serve as entry receptors for certain EV-D68 strains. Lastly, we examined whether EV-D68 strains that bind to HS or glycolipids require different uncoating mechanisms. Bafilomycin A1 minimally affected the cell entry of HS-binding EV-D68 strains B2/039 and B2/947, and the ganglioside preferring B1/2013 and other viruses were strongly inhibited. Together, we identified that EV-D68 strains can use disialoglycolipids as novel receptors and that different EV-D68 strains show a promiscuous sialic acid binding repertoire.

Our study on pasteurising influenza viruses in milk is now out! It's a nice simple story: influenza viruses (including H5N1) are killed really effectively by pasteurisation, but in raw milk they stay infectious - obvious public health implications of both points... (1/2) rdcu.be/d73te

Het afgelopen jaar is het aantal vrouwelijke hoogleraren opnieuw iets gestegen. Ondanks de langzame groei wordt het landelijke streefcijfer van 30 procent aan vrouwelijke hoogleraren volgend jaar waarschijnlijk niet gehaald. Die drempel wordt dit jaar wel voor het eerst gehaald bij de Universitaire Medische Centra UMCs.ppDit jaar is 28,7 procent van de hoogleraren aan Nederlandse universiteiten vrouw. Een groei van 1,1 procentpunt ten opzichte van vorig jaar. Met het huidige groeitempo zal een evenwichtige balans tussen mannelijke en vrouwelijke hoogleraren niet voor 2044 worden behaald. Dat blijkt uit een jaarlijkse monitor van het Landelijk Netwerk Vrouwelijke Hoogleraren LNVH.ph2Carrireladderh2pPer trede op de carrireladder neemt het aandeel vrouwen af, vooral in hogere academische posities, stelt het LNVH. Hoewel meer dan de helft van de studenten en afgestudeerden vrouw is, neemt de vertegenwoordiging van vrouwen af in de daaropvolgende academische niveaus.ppHet aandeel vrouwelijke universitaire docenten lag het afgelopen jaar op 46,9 procent, daarna volgt volgens de monitor een scherpe daling.ph2Erasmus Universiteith2pDertien van de veertien universiteiten zagen vorig jaar een lichte stijging van het aantal vrouwelijke hoogleraren. Alleen op de Erasmus Universiteit daalde het aandeel met 0,2 procent.ppUit de monitor blijkt verder dat in de besturen van universiteiten nu bijna evenveel vrouwen als mannen zitten. Het percentage vrouwen is gestegen naar 48,4 procent. Opvallend genoeg is per 1 november dit jaar de Erasmus Universiteit de eerste Nederlandse universiteit met een geheel vrouwelijk bestuur.ph2Uitstroom mannen en medische faculteitenh2pWederom bevestigt de monitor dit jaar dat de uitstroom van mannelijke hoogleraren in de leeftijdscategorie 60 doorzet. Dit, gecombineerd met de ruim voldoende gevulde talentenpool, biedt uitzonderlijke perspectieven voor de toekomst.ppHet percentage vrouwelijke hoogleraren bij de UMCs is gestegen van 29,8 procent in 2023 naar 31,6 procent in 2024.ph2Donkere wolken boven de sectorh2pDe monitor moet erkennen dat het politieke landschap en het financile plaatje zijn veranderd sinds de publicatie van de vorige editie. Donkere wolken pakken zich samen boven de sector en de voorgestelde bezuinigingen van 1 miljard euro op het hoger onderwijs en onderzoek dreigen werkelijkheid te worden.ppDaarmee doelt de organisatie achter de monitor erop dat diversiteit, inclusie en de focus op gendergelijkheid steeds meer onder druk staan. Mensen in onzekere posities, voornamelijk vrouwen, zijn in het bijzonder kwetsbaar.p
