Postmitotic transcription and 3D regulation show locus-specific and differentiation-specific sensitivity to cohesin depletion - Nature Genetics
Lee et al. deplete the cohesin component RAD21 in mouse embryonic stem cells. Most but not all chromatin loops fail to reform after mitosis, and transcriptional changes were minor, except during diffe...

Coordinated repression of totipotency-associated gene loci by histone methyltransferase EHMT2 via LINE1 regulatory elements - EMBO Reports
Mouse embryonic stem cells (mESCs), in addition to differentiating into the three germ layers, can reverse typical developmental trajectories, as exemplified by their ability to de-differentiate into ...

Excited to share that my most recent work with Kaushiki is now out! See this thread by @stadtfeldlab.bsky.social. As with much of science, the middle of this project felt very murky for a time, before we worked out what was going on. Matthias has hauntingly represented this in his thread.

Very excited to announce that our collaborative study with @efapostolou29.bsky.social on the transcriptional mechanisms distinguishing totipotent and pluripotent cells has been published (link.springer.com/article/10.1...). #totipotency #ZGA #stemcells #epigenetics #chromatin

I'm really happy to announce that my two main postdoctoral works have been published in the same issue of Developmental Cell! First, the final form of the mouse cortical organoid protocol (tinyurl.com/bddfmx8n), which I recently presented at Development presents (tinyurl.com/36udcpme) and…

Our work on the regulation is out www.cell.com/stem-cell-re...! Congrats to our collaborators in the Vierbuchen and Mezey labs. It turns out that genetic variation has to be very much considered if you want to harness stem cells safely in biomedicine. #pluripotency #epigenetics #stemcells

Genetic variation modulates susceptibility to aberrant DNA hypomethylation and imprint deregulation in naive pluripotent stem cells
Pluripotent stem cells are susceptible to acquiring detrimental epigenetic abnormalities, posing a hurdle to their safe use in biomedical applications. Using pluripotent cells with defined genetic bac...

Kaushiki and I are excited to share our preprint about fate restriction in pluripotent cells! We show that EHMT2 counteracts the reactivation of clusters of totipotency genes by nucleating heterochromatin domains at LINE-1 repeats. 1/n @efapostolou29.bsky.social and Stadtfeld Labs #DevBio 🧬🖥️
