Adipocytes: Hidden forces of the immune system?
Zinngrebe et al. propose that adipocyte immunity represents an underappreciated component of host defense, with adipocyte functions extending far beyond energy storage to pathogen sensing and antimicrobial activity. At pathogen entry sites, adipocytes provide local host defense to contain microbial infections and preserve tissue integrity.

Yet, this study scratched barely the surface and further research may understand the precise mechanisms by which shifts in bile acid composition may affect CD8+ T cell immunity directly or indirectly. Moreover, identifying their individual immunomodulatory properties hold its therapeutic potential.

Collectively, we could show an interesting reciprocal crosstalk between CD8+ T cell immunity and systemic bile acid metabolism during viral hepatitis. It further adds to our understanding of how bile acids play an important immunomodulatory role in the body and can change immune outcomes.

Using a genetic knockout model of Slco1a/1b family (Slco-/-), which mediate the hepatic transport of unconjugated bile acids, impaired the CD8+ T cell response, viral clearance and subsequently prevented CD8+ T cell-mediated hepatic damage.

We next asked how shifts in BA levels may impact CD8+ T cell #immunity. Using Myrcludex B, which inhibits NTCP, a hepatic transporter of conjugated bile acids, showed little impact on CD8+ T cell immunity but increased tissue pathology and #hepatocyte damage.

Moreover, we found that LCMV substantially altered gene expression associated with bile acid #metabolism, which was almost exclusively due to CD8+ #Tcells. This was only partially explained by pro-inflammatory #cytokine production or T cell-mediated cytotoxicity.

Liver diseases, including viral #hepatitis, often associate with an increase in systemic bile acid levels and a shift in #bileacid composition. Using the chronic LCMV strain Cl13, we found similar increases and compostional shifts akin to patients - increasing in host-derived conjugated bile acids.
