Screw holes are so hard, especially for reusable ones. I typically use brass fittings that melt into the plastic (using a soldering iron). They work great.

Two preprints up in the same week! This one was from work I finished in 2014, my PhD work! FINALLY. It describes the biochemistry of the yeast glucokinase, which is in the actin family, and forms ATP/ADP-regulated glucose-6-phosphate filaments! www.biorxiv.org/content/10.6...

Love those, today one landed right in front of my, I think trying to catch a gopher. Literally 1 meter away! Once it realized I was there, it flew off quickly, but it was an amazing close encounter I'll never forget.

Forced retirement seems a bit unfair. I hear someone suggest a carrot, some nice grant they can apply for to wind down their career. I looked it up and it was called the Emeritus grant and the NIH never implemented it. I wonder how many people didn’t retire as a result…

I don’t know what the cap should be, but it would be nice to know I have more than a 5% chance of getting a grant when I am applying. A cap may also change rules at certain universities that only let profs be PIs, not to name any specifically…

I personally think capping the number of NIH grants is a good thing - but it would be nice to increase the size of each award a little, considering inflation and stuff. I don’t know what cap is a good cap and how to make exceptions (there should be).

I hundreds of times with BACs and it is mostly rna (I did w/plasmids long ago too). I don’t think rna/dna bits can escape once they are part of the pellet. But I am happy to be proven wrong. Maybe it happens that it is more RNA when there is less DNA (BAC)? Do you use the typical P3 buffer?

This usually makes mostly RNA (my estimation is at least 90% (digested) rna). I do this regularly for BACs where I am just PCRing from it. But if you are loading 90% rna onto the gene gun then you are losing a lot of efficiency.

Its not just the gas, the cheaper maintenance. I only had to fill the wiper fluid and change tires once my car got to 30,000 miles. That is it! And I bought discount tires for less than $100 each.
