The @uni-freiburg.de is once again a University of Excellence!✨ This is a tremendous success for the entire University! At CIBSS, we are proud to have helped lay the foundations for this achievement together with #FutureForests. But something important has already emerged along the way:

🐟 New preprint ! 🐟 For Hippo pathway aficionados. We generated a CRISPR/Cas9 mediated single-copy endogenous Yap1-mGreenLantern fusion protein in medaka to follow Yap1 dynamics in vivo, through embryogenesis and beyond. And it is such a beautiful line ! 🧵

We believe this is just the beginning. Tissue-specific and temporal control of zGrad, combined with our new endogenous KI lines (and those already in the community), will allow us to dynamically interrogate the context-dependent roles of key signalling proteins in development and regeneration.

Ali Seleit @aliseleit.bsky.social · 22/09/2026
Now the fun part: to acutely remove the Yap1 protein in vivo we used the degron nanobody system (zGrad) on our homozygous Yap1-mGL embryos. And it is very potent. Yap1 protein is depleted and we recapitulate the genetic mutant phenotypes in all injected embryos.

Yap1 (hirame) mutants in medaka lose 3D body shape and show wide-ranging phenotypes from the global loss of tissue tension. Our Yap1-mGL homozygotes? Viable, fertile, no phenotype, arguing that the fusion protein is fully functional.

Result: expression and putative activity diverge. High N/C ratio in forming somites and unipotent progenitors in the notochord. Low N/C ratio in posterior PSM and neural tube. Yap1 is everywhere in the tail, but "ON" only in specific tissues.

Ali Seleit @aliseleit.bsky.social · 22/09/2026
Next, we assessed Yap1 signal during somitogenesis. Yap1 activity isn't defined by expression but by sub-cellular (nuclear/cytoplasmic) localisation. So we used the KI line to quantify the N/C ratio in single cells and across the whole tissue as a proxy for Yap1 activity.

Ali Seleit @aliseleit.bsky.social · 22/09/2026
We are able to track the onset of Yap1-mGL fluorescence prior to gastrulation. Cells driving rhythmic contractions during gastrulation show strong Yap1-mGL. Stay tuned for an excellent paper from @simonknoblich.bsky.social & co on the molecular and cellular characterisation of these cells.

It doesn't take a political genius to have seen this coming. The country is sleep-walking into a disaster. The current government is absent/useless. And the political class lost the narrative. A harbinger of dark times.

I think every major company is looking at open weight AI models and realizing they can just bring them in house with an internal engineering team and cut out the frontier model companies to save money. They have more control and privacy. I really don't see frontier labs surviving this.

How about simply reading the work ? We already have working solutions like @reviewcommons.org that include public expert reviews. Coming up with more inherently flawed ways to rank science is a waste of time & effort, it will lead to an amplification of bias hurting scientists & the science.

Hey Anahi...nice work ! Segments are normal sized, so is overall segment number and axis length (when tailbud is amputated). I do think this is an eel-specific feature since the distribution of NMPs is quite unique and the pools are utilized differently over somitogensis.

Ali Seleit @aliseleit.bsky.social · 24/05/2026
Such an important quote you highlight. Drowning in data collection, too little time for actual thought and reflection. Exacerbated by the corporatization of Science. We've created and incentivized a system of fast-food Science production.

Ali Seleit @aliseleit.bsky.social · 24/05/2026
I see ! It is interesting that regeneration is (yet another) module that is dissociable from development. In eels we do not find evidence of regeneration at any stage if the whole PSM is amputated but they are able to continue segmenting after tailbud amputation (likely using their rostral NMPs)

Ali Seleit @aliseleit.bsky.social · 24/05/2026
Nice work Wouter and team ! Just a methodological question when you amputate the tailbud you mean the whole unsegmented PSM until the last formed somite correct ?

Ali Seleit @aliseleit.bsky.social · 10/05/2026
I think that might very well be ! We did not find evidence of any 'traditional' posterior gradient in the eel PSM... interestingly the core clock pathway your lab specializes in: her->dusp->ppERK(protein) is conserved (and dynamic!)...the wave itself seems to be pulsatile (ON/OFF) in the eel PSM
