5/ While the exact mechanism of sodium pumping remains unknown, we hypothesise the metallocluster acts as the Lewis acid activating CoM. Sodium ion binding at the conserved site may be destabilised by CoM deprotonation putatively propagated via Asn and Asp residues, triggering ion translocation.

4/ MtrA binding to MtrCDE and corrinoid insertion also correlate with rearrangements within the MtrCDE membrane plane. MtrD 48–68 undergoes the largest shift, a movement at the MtrDE interface that widens a potential sodium ion translocation pathway.

3/ Binding and insertion of the corrinoid into the MtrCDE vestibule correlates with changes of CoM binding. Although not unambiguously assignable, an elongated feature appears at the apical iron of the cluster, implicating the clusters direct participation in methyl transfer.
