How do lysosomes rapidly adapt to membrane tension to avoid bursting? In this article, @laylanassar.bsky.social and I highlight discoveries from Angela Kim, Spencer Freeman and colleagues who show a role for TMEM63A, a mechanosensitive cation channel in protecting lysosomes doi.org/10.1083/jcb....

We’ve engineered stable cell systems to induce lipid scrambling of the plasma membrane with active forms of TMEM16F. Scrambling alone causes vesiculation (🔵TMEM16F-active; 🟠PS/Annexin V). Let us know if you want to try the cells out. Preprint: www.biorxiv.org/content/10.1...
HeLa cells stably expressing a dox-inducible active TMEM16F-GFP (blue). Once expressed, lipid scrambling of the plasma membrane bilayer causes vesiculation, shown with Annexin V staining of the exposed PS (gold).

Sorting nexin 10 regulates lysosomal ionic homeostasis via ClC-7 by controlling PI(3,5)P2
Snx10 was found to determine the level of PI(3,5)P2 in (phago)lysosomes by controlling the supply of PI(3)P from upstream endocytic compartments. By regula

We've been studying mechanisms of resilience that protect lysosomes from damage and wrote a review on the topic. We consider lysosomal stress and damage on a spectrum. Please help me share as this is my first post 😀 and I don't have many followers! 🙏 Open Access: www.embopress.org/doi/full/10....
