New preprint from the Gurney Lab, on bioRxiv: The Phage Time Continuum: Genome Organization Constrains Anti-CRISPR Success. www.biorxiv.org/content/10.6... I think this is one of the cooler things I'll ever do. Work lead by @sczerwinski.bsky.social he coined the term Phage Time Continuum. 1/10

The Phage Time Continuum: Genome Organization Constrains Anti-CRISPR Success
Bacteria and lytic phages experience antagonistic co-evolution, with bacterial defenses (e.g., CRISPR-Cas) battling phage anti-defenses (e.g., anti-CRISPR (Acr) systems). We investigate whether CRISPR...

How can a #GiantVirus 👾 lurk inside the host genome undetected? We find that epigenetic conflict lies at the centre of the ⚖️ between infection and co-existence with the host. Great collaboration with the Aylward lab out in @biorxivpreprint.bsky.social: www.biorxiv.org/content/10.6...

Most animal species pack sperm DNA with protamines instead of histones. Protamines are essential for male fertility. They are among the fastest-evolving genes in animals. That makes little sense. We think we know why, in a new paper led by @ching-hochang.bsky.social. www.science.org/doi/10.1126/...

🚨🚨Preprint alert!🚨🚨 We removed the main centromere 3 domain of Drosophila‼️ We designed an inducible dCas9-Fok1 system to cut 🧬 at unique sequences flanking the ~50-kb island where most of CENP-A sits. To our knowledge, this has never been attempted in a living animal!

Direct cell-to-cell transmission of retrotransposons
Retrotransposons can infect neighboring cells using a mechanism that relies on a small fusogen-like protein rather than on the envelope protein that is typically required for infection.

Discovery #1: The mechanism. These retrotransposons transmit directly between cells using fusogen-like sORF2 proteins. 🔑 sORF2 drives invasive cellular protrusions that physically reach the oocyte👉, and deliver retrotransposon capsids without an extracellular step. (4/7)

Why was this such a surprise? Until now, the only transposons known to transmit between cells were those carrying an Envelope protein (like endogenous retroviruses). The elements we studied completely lack an Envelope - so how were they crossing cellular boundaries? 🤔 (3/7)

Co-option of retrotransposons promotes antibody diversification
A longstanding mystery in antibody diversification is how AID specifically targets immunoglobulin V genes. Ancient LINE-1 retrotransposons embedded within the immunoglobulin locus provide a recruitment platform for the HUSH complex and AID to drive somatic hypermutation.
