A cornerstone discovery from our lab is that cGAMP, a second messenger, can transmit between cells to broadcast danger and coordinate an immune response. In our new preprint, we show that its role as an immunotransmitter is detrimental for neuroinflammation. www.biorxiv.org/content/10.6...

Extracellular cGAMP transmission exacerbates neuroinflammation in aging and multiple sclerosis
Neuroinflammation drives neurodegeneration and cognitive decline in aging, but the signals that spread this inflammation across the CNS remain unclear. Here, we show in mice that advanced age and expe...

In the latest published work from the lab, we show that toxicity in T cells in therapies using STING agonists is due to import by SLC7A1, offering a new therapeutic avenue that complements current approaches that harness innate immunity in cancer therapy. www.cell.com/cell-reports...

ENPP1 inhibitor with ultralong drug-target residence time as an innate immune checkpoint blockade cancer therapy
Tumor-selective ENPP1 inhibitor is highly sought after for cancer immunotherapy to activate anti-cancer innate immunity without systemic inflammation. Wang et al. achieved this with STF-1623, which ha...

Activation of the stimulator of interferon genes (STING) pathway can be great for fighting pathogens and treating cancer. But when the pathway is too active, it can do more harm than good, aggravating autoimmune and neurodegenerative diseases. cen.acs.org/analytical-c... #chemsky 🧪
