Tend to agree. Work by us & @kdmicheva.bsky.social showed that a strangely high fraction (~50%) of myelin in upper layers of cortex wraps short lengths of fast-spiking, parvalbumin-positive basket cells. We suggested it was to support their axons, rather than to speed up spike time arrival.

The GeneTex GTX13970 works well for me, on glutaraldehyde fixed tissue as well. For TdTomato: Origene AB8181. Both used postembedding, Lowicryl HM20

We should definitely think and talk more about this! Ignoring these blind spots can be damaging to both our science and our morality. Some of us try to appease our conscience by sparing other animal lives - gently taking spiders outside, becoming vegetarian, but that does not resolve the conflict

Looks like autofluorescence to me (not an olfactory expert). Could it be lipofuscin, I see that a lot in cortex where I mostly look? A quick search of lipofuscin and olfactory bulb brought this paper and the images look pretty similar to yours: pubmed.ncbi.nlm.nih.gov/19666062/

Our paper is out just in time for #EM_Monday! It is about a specialized neuron-specific ER, called the spine apparatus, which is found close to synapses and has a very peculiar shape. Thank you! This is one way of looking inside biological tissue, where everything is so tiny and crammed. Just slice it up real thin and that's what this diamond knife and ultramicrotome are used for. After this, we'll label it in different ways and look at the tissue slices with a microscope

This is a gorgeous image! The myelin is so graceful at this magnification! It does look like there is not that much myelin in cortex, in comparison to the rest, but when you zoom in, the way I look at it, there's still a lot. Look at layer 5 (white is MBP, red is GABA):
Mouse cortex imaged with array tomography. There is a red neuron in the middle that is stained with a GABA antibody. A dozen of blue nuclei are surrounding it, and myelinated axons looking like white tubes are interwoven all around them. And all of this is sprinkled with red dots, the inhibitory synaptic boutons labeled with GABA.

Time for some of my favorite images with immunofluorescence array tomography: this one is human cortex, and the yellow fluff around the blue nuclei are lipofuscin granules that accumulate with aging. Lipofuscin in this image comes for free - no label needed because it is autofluorescent!
