Happy to share some of the auxin Marchantia research we've been working on for a while with @fromani.bsky.social and @jimhaseloff.bsky.social. We confirm long standing suspicions that stem cells in Marchantia exist in an auxin concentration/signalling minimum, and potentially a cytokinin maximum.

Fig. 9 (shortened, full legend in paper): Summary and model for the role of N-hydroxy pipecolic acid (NHP) and salicylic acid (SA) in Arabidopsis. SA is synthesized via phenylalanine (Phe) and isochorismic acid (ICA) using several enzymatic steps (black arrows). NHP is synthesized from the precursor lysine (Lys) also by different enzymatic steps symbolized and via pipecolic acid (Pip) via one enzymatic step, the protein FLAVIN-DEPENDENT MONOOXYGENASE 1 (FMO1). Both SA and NHP are conjugated to inactive signals using a glucosyltransferase reaction (UGT76B). In this study we used mutation in the gene that results in up-regulation of SA and also NHP. The same phenotype can be achieved by overexpression of FMO1 in a dominant mutation (fmo1D). Knockout of FMO1 leads to reduction of NHP. Plasmodiophora brassicae has an effector protein PbBSMT capable of methylating both SA and NHP in vitro (orange arrows).
