A recent @cp-cell.bsky.social paper proposed that double-stranded DNA directly binds and activates SARM1. In a new @pnas.org paper, an international team was unable to reproduce that central finding www.pnas.org/doi/abs/10.1...

AlphaFold Server’s ligand menu just got a lot bigger and I almost missed it! You can now add any ligand from the wwPDB Chemical Component Dictionary. Just enter its CCD code 🥳

A massive update: At least ~15 companies~ are selling scientists antibodies using faked validation data. We've documented 18,000+ manipulated images on 17,000+ products sold by leading laboratory suppliers including Thermo Fisher, Abcam, Santa Cruz Biotechnology, Millipore Sigma and Bio-Techne. 1/🧵

Today I'm posting a new blog! Nearly two hundred papers have muddled the names for different proteins and recorded using the wrong antibody for a range of experiments. This time it's PERK vs. p-ERK. Including researchers from Harvard, the Mayo Clinic, and more. sholto.substack.com/p/perk-vs-p-...

I’ve officially resigned as Associate Editor for Frontiers in Systems Neuroscience. It used to be a reputable journal, but became a case study in how forced automation destroys academic integrity. 👇

Thermo Fisher now has a page up in response to our observations. They flatly deny that images were manipulated or fabricated while repeating that images were "optimized for presentation and clarity on the website", no lie, six times. www.thermofisher.com/us/en/home/l...

Surprised to discover that Thermo Fisher appears to show a fake western blot for the validation of one of their p53 antibodies. I've added a diagram to show the very similar bands. This does not appear to be one of the "published figures" but their own internal data.

If I claimed this liquid-to-solid phase transition assay on a pure protein in vitro predicts the same transition to happen in cells, what counterarguments would you raise? Would the same counterarguments apply to a liquid-to-liquid phase transition assay on the same pure protein in vitro?

Neat trick if you polycolonal ab's suck. Incubate them with fixed cells with a KO of your protein of interest, then spin. Protocol here: www.med.upenn.edu/markslab/ass... I was amazed how well it worked on first try (I'm sure that I can completely eliminate unspecific bands) #WesternBlot #cellsky
On the left - western blot of B16F10 cells wt and KO for CDK8. Our in house produced antibodies give a lot of unspecific bands. On the right same probes with antibodies preincubated with fixed CDK8 KO cells - there is a specific band and faint unspecific bands, which can be probably eliminated with increase of amount of KO cells.

Peer review has a weakness in that it is not in one's interest to upset colleagues by pointing out their mistakes, as upset peers may write negative reviews in the future. But setting the record straight is an essential part of science, and an ever-self-congratulating field is prone to get stuck.
