Interestingly, the same mechanism is found in mice: the mutually antagonistic interaction between postsynaptic proteins SRGAP2A and SYNGAP1, that sets the tempo of synaptic maturation, is conserved in mammals. Therefore, SRGAP2B/C are human modifiers of this ancestral mechanisms.

But whether and how SRGAP2B/C play a critical role in synaptic neoteny, remained to be explored. Using our xenotransplantation model, SRGAP2B/C loss-of-function induced an acceleration of the synaptic maturation. After 18 months, densities are similar to those observed in 5/10-years-old humans!

Previous studies showed that human cortical pyramidal neurons develop along a prolonged neotenic timescale when xenotransplanted in the fast-developing mouse cortex, that we confirmed for the synaptic development itself. These results point to species-specific, cell-intrinsic, mechanisms.
